Three things to check: make sure you're connected to your office VPN, make sure you have the correct computer name, and make sure the office computer hasn't gone to sleep or locked. If you're on VPN and have the right name, the computer on your end is probably sleeping.
Remote Desktop only works when you're connected to your office network, either through a VPN or a direct company connection. If the connection fails, you're either not on the VPN, using the wrong computer name, or the office computer is asleep. Start with VPN and computer name, then wake up the office computer if needed.
